胞子抽出

1. １Lホーロービーカーに土壌を少量取る。take a small amount of soil in a 1L beaker
2. １Lビーカーに少量の水道水を加え、土壌を手でよく揉みほぐす。add small amount of tap water and disperse the soil well by hands
3. １Lビーカーに水道水を勢いよく注ぐ。pore tap watervigorously
4. 10秒間静置したのち、上清を篩いに通す。place 10 s and pass the supernatant through sieves
5. 106 µmメッシュ篩い上に残った土壌を水道水で集める。gather spores on the corner of the sieve with 106 µm mesh usning tap water
6. 洗瓶を用いて篩上の粒子を集め、小ビーカー（50-100ml）に流し込む。take spores in a beaker using water
7. 小ビーカーを20秒間超音波洗浄器にかける。sonicate for 20 s
8. 小ビーカーの土壌をもう一度篩いに通す。pass them through the sieves
9. 106 µmメッシュ篩い上に残った土壌を小ビーカーに移す。take spore on the sieve into the beaker
10. 一部をシャーレにとり、胞子用のピンセットを用いて実体顕微鏡下で胞子を拾う。move a portion of spores into a petri dish and take spores under dissect microscope using flexible forceps
11. 蒸留水が入った小シャーレに胞子を入れる。add spores into a small dish with pure water
