菌糸蛍光染色

1. ファーマー液（氷酢酸：エタノール＝１：３）で固定してある場合には、純水で数分間×数回洗浄する。wash with pure water several min several times, if roots are stored in farmer's solution (acetate:ethanol=1:3)
2. 根を10% KOH中で90℃10分間浸漬する。incubate roots in 10% KOH solution at 90℃ for 10 min using a water bath.
3. 水で2-3回リンスする。
4. 0.1 M HClで30分間インキュベートする。
5. PBSで根を数回洗浄する。wash with PBS several times.
6. 脱気する。(やらなくて良い) degas, if necessary.
7. 根を5 µg/mlのWGA-Oregon green 488（PBS）で１15分〜数時間遮光して染色する。（回転させながら染色） stain roots with 5 µg/mlWGA-Oregon green 488in PBS for several hours in dark with rotation.
8. PBSで根を数回洗浄する。wash roots with PBS several times.
9. 根を5 µg/mlのpropidium iodide（PBS）で30分間染色する。stain roots with 5 µg/ml propidium iodide in PBS for 30 min, if necessary.
10. PBSで根を数回洗浄する。(7,8はやらなくても良い) wash with PBS several times.
11. プレパラートを作製し(純水を滴下し、マニキュアで封をする)、蛍光顕微鏡で観察する。mount roots with pure water
and observe using a fluorescent microscope.
